Police confiscated the plants after executing a search warrant at a home on Maya Drive, Medowie about 8.30am on Thursday.
A total of 305 mature plants and seedlings with an estimated potential street value of more than $600,000 as well as drug paraphernalia, were seized, police said.
The raid comes after the Port Stephens-Hunter Police District Proactive Crime Team commenced an investigation into the cultivation of cannabis in the Medowie area earlier this month.
Investigations are continuing into the cultivation of the plants.
