Wells' parents, Kylie and Darren, and younger sister Emma were huddled around the television in their family home, joined via Facetime by his twin brother Lachlan.
Godfather and former coach Shane Barclay also caught the milestone moment live.
Old teammates from Miller Field and club president Stephen Pack did likewise.
The 24-year-old achieved a lifelong dream and became the 36th Australian to play MLB when he was called up by the Baltimore Orioles against the Toronto Blue Jays at Sahlen Field.
It was the bottom of the seventh and Wells, a left-handed pitcher, made his way to the mound.
"It was very emotional and we were all very proud," Wells' mum Kylie told the Newcastle Herald.
"It was a shame we couldn't get over there because of COVID-19, but we were all watching together.
"We spoke to him after the game and told him how much we loved him."
Barclay said Wells "really works hard and deserves everything that comes his way".
Pack, the head of Belmont Baseball Club where Wells started his career as a junior, said to "have someone from our club representing on the world stage in MLB is just amazing".
Wells had two scoreless innings with Santiago Espinal his first out. He also dismissed Blue Jays gun Vladimir Guerrero jnr.
An MLB.com reporter described Wells' maiden run as "encouraging" and "one of the few silver linings" from Baltimore's 12-4 loss.
Wells, unused on Saturday after being promoted from triple-A side Norfolk, could have game four with the Orioles on Monday (AEST). Toronto leads the series 2-1.
Ryan Rowland-Smith, a former Newcastle High School student, made his MLB debut with the Seattle Mariners in 2007.
