Vice-Chancellor Alex Zelinsky issued a statement to staff via email about 8.20am on Saturday.
NSW Health has transferred the students into "care", he said.
"It is with concern that I advise you that two of our students who are staying in our on-campus accommodation at Callaghan have tested positive to COVID-19," Dr Zelinsky said.
"Several close contacts of the two students at the student residence have been identified by NSW Health, and these students have been contacted, tested, and moved into self-isolation.
"NSW Health and the University are staying in close contact with this group of students to monitor their health and support their wellbeing during their period of isolation."
Dr Zelinsky said the news would be "unsettling" but he advised all students and staff to stay vigilant.
"It is a real reminder that COVID-19 is active in our community," he said.
"Therefore, I ask you to be highly vigilant for signs of COVID-19,and if symptoms emerge, immediately contact health.response@newcastle.edu.au, get yourself tested, and then self-isolate until a negative result is obtained.
"Please continue to strictly follow the stay-at-home orders in place for our region. These measures are there to keep all of us safe and to reduce our risk of exposure."
